Abstract
目的:探讨 α1A 肾上腺素受体在阿尔茨海默病(AD)中枢神经炎症中的作用。方法:通过生物信息学分 析 α1A 肾上腺素受体在野生型(WT)小鼠和 3xTg-AD 小鼠模型中的表达及作用;通过 RT-qPCR、Western blot 和免 疫荧光检测小鼠脑中 α1A 肾上腺素受体的表达和定位;给予小鼠 α1A 肾上腺素受体阻断剂哌唑嗪(Prazosin)处理, Western blot、免疫荧光染色检测小鼠脑部 iNOS 和 CD68 的表达;RT-qPCR 和 ELISA 分析炎性细胞因子 IL-1β、 IL-6、TNF-α 的表达水平。结果:α1A 受体在 3xTg-AD 小鼠的小胶质细胞上调;阻断 α1A 受体能减少小胶质细胞的活 化以及炎症因子 IL-1β、IL-6 和 TNF-α 的释放。结论:α1A 肾上腺素能受体在 AD 脑部上调,并引起中枢炎症。
